Resumen
The sugar maple is a native species of northeastern North America and is widely appreciated and exploited for its sap, which is concentrated to produce maple syrup. However, its bark is not widely used, despite its content of a variety of bioactive compounds which could be applied in health products. Apart from polyphenols, higher molecular weight compounds such as oligo- and polysaccharides have been less explored so far. The aim of this investigation was to isolate and characterize a complex sugar from sugar maple bark and evaluate its biological activities. Two samples obtained by extraction with water and ethanol-water (v/v:1/1) at 80°C, have been studied in this research. Sugar components were concentrated by ethanol (95%) precipitation and ultrafiltration (5KDa MWCO) to obtain enriched polysaccharides fractions. The latter were separated by gel filtration and anion exchange chromatography. Among the bark maple polysaccharides, this study was focused on one of the neutral polysaccharides, inulin detected in maple syrup. Inulin has a well documented prebiotic activity. Monosaccharide compositions, linkage analyses, molecular weight and degree of crystallinity of the isolated polysaccharides were investigated by NMR, GC-MS, gel permeation chromatography (GPC) and X-ray diffraction. Biological studies will be performed on crude extract samples, sugar enriched samples and isolated complex polysaccharide.
